Large sequence-defined superstructures containing several hundreds of monomers were prepared by a combination of stepwise solid-phase chemistry and DNA-guided self-assembly. Bio-hybrid precursors containing synthetic sequence-defined segments connected to short single-stranded DNA extremities were first synthesized by automated phosphoramidite chemistry. Afterwards, the sequence-specific hybridization of the DNA segments enabled preparation of supramolecular topologies with controlled sizes and shapes. These results open unprecedented avenues for the development of molecularly-defined 2D and 3D polymer architectures.
